London: A team of researchers, led by those of Indian-origin, has in a breakthrough developed a single gene therapy injection that could dramatically reduce the bleeding risk faced by people with haemophilia B.
Haemophilia B is a rare and inherited genetic bleeding disorder caused by low levels of the factor IX (FIX) protein, needed for forming blood clots that help prevent or stop bleeding.
The gene responsible for making FIX protein is located on the X chromosome, so the severe form of haemophilia B is much more common in men.
Currently, patients with haemophilia B need to inject themselves regularly usually weekly with recombinant FIX, that is, regular replacement therapy to prevent excessive bleeding. Despite advances in treatment, patients may continue to see debilitating joint damage.
But the new type of adeno-associated virus (AAV) gene therapy candidate called FLT180a, developed by researchers from University College London treats severe and moderately severe cases of the condition.
The study, published in the New England Journal of Medicine, showed that one-time treatment with FLT180a led to sustained production of FIX protein from the liver in nine of 10 patients across four different dose levels, removing the need for regular replacement therapy.
"Removing the need for haemophilia patients to regularly inject themselves with the missing protein is an important step in improving their quality of life," said lead author Professor Pratima Chowdary, Royal Free Hospital at UCL Cancer Institute.
AAV gene therapy works by using a packaging from the proteins found in the outer coat of the virus, to deliver a functional copy of a gene directly to patient tissues to compensate for one that is not working properly.
Newly synthesised proteins are released into the blood and a one-time infusion can achieve long-lasting effects.
Patients needed to take immunosuppressive drugs over several weeks to several months to prevent their immune systems from rejecting the therapy.
While the treatment was generally well tolerated, all patients experienced some form of adverse events, with an abnormal blood clot in one who received the highest FLT180a dose and had the highest levels of FIX protein.
In nine out of the 10 patients, the treatment led to a sustained increase in FIX protein production, which led to a decrease in excessive bleeding. They also no longer required weekly injections of FIX protein.
After 26 weeks, five patients had normal levels of FIX protein, three had low but increased levels, and one patient treated at the highest dose had an abnormally high level. (Agency)

London: Finding it hard to raise levels of Vitamin D, popularly known as the sunshine vitamin?
Scientists have developed gene-edited tomatoes that could be a simple and sustainable innovation to address the global health problem.
Researchers John Innes Centre in the UK used gene editing to turn off a specific molecule in the plant's genome which increased provitamin D3 in both the fruit and leaves of tomato plants. It was then converted to Vitamin D3 through exposure to Ultraviolet B light.
Vitamin D is created in our bodies after skin's exposure to UVB light, but the major source is food.
This new biofortified crop could help millions of people with Vitamin D insufficiency, a growing issue linked to higher risk of cancer, dementia, and many leading causes of mortality, according to the study, which appears in the journal Nature Plants.
Tomatoes naturally contain one of the building blocks of Vitamin D3, called provitamin D3 or 7-dehydrocholesterol (7-DHC), in their leaves at very low levels. Provitamin D3, does not normally accumulate in ripe tomato fruits.
Researchers used CRISPR-Cas9 gene editing to make revisions to the genetic code of tomato plants so that provitamin D3 accumulates in the tomato fruit. The leaves of the edited plants contained up to 600 microgram of provitamin D3 per gram of dry weight. The recommended daily intake of vitamin d is 10 micrograms for adults.
When growing tomatoes leaves are usually waste material, but those of the edited plants could be used for the manufacture of vegan-friendly Vitamin D3 supplements, or for food fortification.
"We've shown that you can biofortify tomatoes with provitamin D3 using gene editing, which means tomatoes could be developed as a plant-based, sustainable source of vitamin D3," said Professor Cathie Martin, corresponding author of the
"Forty per cent of Europeans have vitamin D insufficiency and so do one billion people world-wide. We are not only addressing a huge health problem, but are helping producers, because tomato leaves which currently go to waste, could be used to make supplements from the gene-edited lines," Martin added.
The study noted that the provitamin D enriched tomatoes could be much helpful for people adopting a plant-rich, vegetarian or vegan diet, and for the growing number of people worldwide suffering from the problem of vitamin D insufficiency. (Agency)

Patna: The health department of Bihar on Thursday detected a new variant of Omicron in Indira Gandhi Institute of Medical Sciences (IGIMS).
The new variant BA.12 is 10 times more dangerous than BA.2 which was detected during the third wave of Corona in the country.
Prof Dr Namrata Kumari, the HOD of microbiology department of IGIMS, said: "Keeping in view the rising Covid cases, we had started genome sequencing of samples of Omicron variant of Corona. There were 13 samples tested and one of them had BA.12 strains. The remaining 12 samples have BA.2 strains."
"We have asked the authority for contact tracing of all the positive samples of Omicron. The BA.12 variant is 10 time more dangerous than BA.2. Though, there is no need to worry. Precaution is required here to protect from it," She said.
The BA.12 variant was first detected in the US. There were two to three cases detected in Delhi and now one case in Patna. (agency)

Lucknow: Students at the Lucknow University (LU) will now have a new option for an under-graduate course in Naturopathic and Yogic Science (BNYS) from the academic session 2022-23.
According to LU spokesperson Durgesh Srivastava, the duration of the course will be 5.5 years. Candidates can apply for admission on 60 seats of the course that will be offered under LU's Institute of Yogic Studies.
"The application process for the course is the same as other UG courses. Students will have to apply online on LU's official website. The last date to apply for admission is May 31," he said and added that a candidate should have at least 50 per cent marks in biology in Class 12 to be eligible to apply for admission in the course.
The admission will be on the basis of the merit of entrance test.
Students, who are appearing for the intermediate exam this year, can also apply and appear in the entrance exam. The age of the student should not be less than 17 years at the time of admission. The five years of the course will include one year of mandatory internship."
Srivastava said the BNYS course is related to medical science and has high employability.
In this, the study of body structure, body mechanism, changes in the body during disease and natural methods of identifying diseases such as facial morphology and others will be done.
