An Ayurvedic poly-herbal formulation, NEERI-KFT, has the potential to not only slow down the progression of chronic kidney disease but also restore normalcy in functional parameters of the vital organ, a team of researchers has claimed.
In a review published in the latest edition of Saudi Journal of Biological Sciences, they said "phytopharmacological evaluation of NEERI-KFT suggests that it exhibits substantial potential against oxidative and inflammatory stress induced apoptosis by exerting antioxidant, nephroprotective and immunomodulatory effects...in the patients associated with renal dysfunction or chronic kidney disease (CKD)".
The NEERI-KFT, a herbal medicine extracted from plants, has been found to correct oxidative as well as inflammatory stress known for reducing body's immunity, said the authors after drawing data on the formulation for kidney ailments from more than five electronic databases such as Science Direct, Google Scholar, Elsevier, PubMed, Springer, ACS publication from published database between 2000 and 2020.

The study, 'A systematic review on nephroprotective AYUSH drugs as constituents of NEERI-KFT' not only examined the impact of the herbal extract of NEERI-KFT prepared by the AIMIL Pharma interventions on patients, but also dwelled in detail the adverse events of the modern medicine used for the management of the kidney ailments.
The study also highlighted the herbal drug's efficacy in reducing "serum creatinine, blood urea, and serum uric acid as compared to placebo group" and described "its well-tolerated effect with no adverse hematological or biochemical abnormalities occurred to any subjects during clinical trial".
"It can be suggested that NEERI-KFT can be an alternative and complementary therapy at end-stage renal disease (ESRD) or complete loss of kidney function where frequent dialysis of renal transplant remains only a single option for continued survival CKD patients," researchers said.
K.K. Sharma, AIMIL Pharma Managing Director, attributed this to the "more than 20 different potent herbs like haridra, varuna, shirish, gokhru, punarnava and anantamul to name a few in the formulation which are known for their nephro corrective, anti-inflammatory and antioxidant properties and reparative regeneration of kidney cells.

It was after a string of stringent tests that this formulation has been developed to provide relief to kidney patients, he said.
K.N. Dwivedi from Banaras Hindu University said the herbal ingredients in NEERI-KFT are loaded with many anti-oxidants besides medicinal values which not only strengthen kidney but liver also.
"This is a very unique drug and we have found it very effective in our study also," he added.

New York -The airway cells of patients with chronic lung diseases are "primed" for infection by the Covid-19 virus, resulting in more severe symptoms, poorer outcomes and a greater likelihood of death, according to a study.
The study, published in Nature Communications, found that chronic lung disease causes genetic changes in the molecular makeup of a variety of cells, including the epithelial cells that line the lung and airways.
The changes enable SARS-CoV-2, the virus that causes Covid-19, to enter the body, replicate and trigger an out-of-control immune response that fills the lungs with fluids and often results in patients needing respirators and lengthy hospitalisations.
"Our results suggest that patients with chronic lung disease are molecularly primed to be more susceptible to infection by SARS-CoV-2," said Nicholas Banovich, Associate Professor at Translational Genomics Research Institute, a non-profit genomics research institute in Arizona, US.
In addition, older-age, male-gender, smoking, and comorbidities such as high blood pressure, obesity and diabetes, are all Covid-19 risk factors that are exacerbated by chronic lung diseases, such as Chronic Obstructive Pulmonary Disease, Interstitial Lung Disease, and especially Idiopathic Pulmonary Fibrosis, a progressive scarring and stiffening of the lung tissue.
For the study, the team used single-cell RNA sequencing technology to spell out the genetic code of 611,398 cells from various databases, representing those with both healthy (control) lungs and those with chronic lung disease. Sequencing and analysis allowed researchers to identify molecular characteristics that may account for worse Covid-19 outcomes.
Researchers specifically searched for changes in AT2 cells -- a major lung epithelial cell type, focusing on cellular pathways and expression levels of genes associated with Covid-19. They established a "viral entry score," a composite of all genes associated with SARS-CoV-2, and found higher scores among cells from patients with chronic lung disease.
Further, exploring changes in immune cells, they discovered dysregulated gene expression associated with hyper-inflammation and with sustained cytokine production -- two signature symptoms of severe SARS-CoV-2 infection. So-called cytokine storms in Covid-19 patients unleash a cascade of immune cells that flood the lungs, causing severe organ damage, the team explained. (Agency)

New York, May 20 (IANS) Young adults who survived Covid-19, even with mild infections, are at increased risk of a serious post-Covid or "long haul" problem -- chronic fatigue syndrome, say researchers.A recent review of three case studies by Johns Hopkins University researchers provides first evidence that one serious post-Covid problem may be myalgic encephalomyelitis/ chronic fatigue syndrome (ME/CFS) -- the complex, multisystem disorder previously known as chronic fatigue syndrome."In the three patients studied -- all of whom had confirmed or highly probable Covid-19 infections early in the pandemic -- we observed ME/CFS-like symptoms within the first two weeks of illness," said Peter Rowe, director at Johns Hopkins Children's Center and Professor of Paediatrics at the Johns Hopkins University School of Medicine."At six months following their illness, all three still met the criteria for being diagnosed with ME/CFS," Rowe added.The three patients evaluated in the recent study, published in the journal Frontiers in Medicine, were a 19-year-old man and two women, ages 22 and 30, whose Covid-19 symptoms began between April and June 2020.Symptoms of orthostatic intolerance -- a group of clinical conditions that includes fatigue, lightheadedness and difficulty concentrating, and are linked with greater than 90 per cent of the people with ME/CFS -- were prominent in all three from the outset of their Covid-19 illness.A six-month post-Covid symptom onset examination, including evaluations of movement, neurological function and continued orthostatic intolerance, was conducted on each of the patients to determine if ME/CFS could be diagnosed. New York, April 3 (IANS) Researchers have identified the biological mechanism by which chronic stress impairs hair follicle stem cells, confirming long-standing observations that stress might lead to hair loss.
In a mouse study, published in the journal Nature, the researchers found that a major stress hormone puts hair follicle stem cells into an extended resting phase, without regenerating the follicle or the hair.A
The researchers identified the specific cell type and molecule responsible for relaying the stress signal to the stem cells and showed that this pathway can be potentially targeted to restore hair growth.
"The skin offers a tractable and accessible system to study this important problem in depth, and in this work, we found that stress does actually delay stem-cell activation and fundamentally changes how frequently hair follicle stem cells regenerate tissues," said researcher Ya-Chieh Hsu from Harvard University.
The hair follicle naturally cycles between growth and rest, a process fuelled by hair follicle stem cells. During the growth phase, hair follicle stem cells become activated to regenerate the follicle and hair, and hairs grow longer each day.
During the resting phase, the stem cells are quiescent and hairs shed more easily. Hair loss can occur if the hairs shed and the stem cells remain quiescent without regenerating new tissue.
The researchers studied a mouse model of chronic stress and found that hair follicle stem cells stayed in a resting phase for a very long time without regenerating tissues.A
A major stress hormone produced by the adrenal glands, corticosterone, was upregulated by chronic stress; giving mice corticosterone reproduced the stress effect on the stem cells.A
The equivalent hormone in humans is cortisol, which is often referred to as the "stress hormone."
Under normal conditions, hair follicle regeneration slows over time -- the resting phase becomes longer as the animals age.
But when the researchers removed the stress hormones, the stem cells' resting phase became extremely short and the mice constantly entered the growth phase to regenerate hair follicles throughout their life, even when they were old.

However, the researchers, including Jochen Lennerz from Massachusetts General Hospital, have also found a way to block the pathway to cold-sensitive teeth.
"We found that odontoblasts, which support the shape of the tooth, are also responsible for sensing cold," said Lennerz.
Teeth that hurt from exposure to cold can occur for many reasons. Many people have experienced intense pain from cold when they have a hole in a tooth from an untreated cavity.
For the study, the team conducted experiments on mice whose molars were drilled under anesthesia.
Mice with dental injuries manifest pain with their behaviour; they drink up to 300 per cent more sugar water than their litter mates without dental injuries, for example.
In previous research, the team of investigators had discovered TRCP5, a protein encoded by the TRCP5 gene that is expressed in nerves in many parts of the body. Their earlier discovery allowed the researchers to zero in on TRCP5 as a mediator of pain from cold.
By studying genetically altered mice that did not have the TRCP5 gene, the researchers found that the mice with injured teeth did not manifest the increased drinking behaviour and behaved like mice without dental injuries.
The research team also identified a pharmacological target for minimising tooth sensitivity to cold. For centuries, oil of cloves has been used as a remedy for tooth pain. The active agent in oil of cloves is eugenol, which happens to block TRCP5. (IANS)
