Tokyo, July 2 (IANS) A team of scientists has identified an anti-malarial drug, mefloquine, that is effective against SARS-CoV-2, the virus causing Covid-19.The team from Tokyo University of Science and others mathematically modelled the effectiveness of mefloquine to predict its potential real-world impact, if applied to treat Covid-19.They found that mefloquine could reduce the overall viral load in affected patients to under 7 per cent and shorten the "time-till-virus-elimination" by 6.1 days. Their findings are published in Frontiers in Microbiology.To identify drugs with higher antiviral potency than existing antivirals, the team first screened approved anti-parasitic/anti-protozoal drugs.They found that mefloquine had the highest anti-SARS-CoV-2 activity among the tested compounds. Upon testing it against other quinoline derivatives, such as hydrochloroquine, in a cell line mimicking the cell-based environments of human lung cells, they found it to be better."In our cell assays, mefloquine readily reduced the viral RNA levels when applied at the viral entry phase but showed no activity during virus-cell attachment. This shows that mefloquine is effective on SARS-COV-2 entry into cells after attachment on cell surface," said lead scientist Koichi Watashi, from the varsity.To bolster mefloquine's antiviral activity, the scientists looked into the possibility of combining it with a drug that inhibits the replication step of SARS-CoV-2: Nelfinavir.Interestingly, they observed that the two drugs acted in "synergy" and the drug combination showed greater antiviral activity than either showed alone, without being toxic to the cells in the cell lines themselves.While the study must be succeeded by clinical trials, "the world can hope that mefloquine becomes a drug used to effectively treat patients with Covid-19," the researchers noted.--IANSrvt/vd
Hyderabad, July 1 (IANS) Amid the rush to develop cures for the coronavirus, a study by a University of Hyderabad (UoH) incubated startup, has discovered that an anti-diabetic drug has potential use as a ready to use, cost-effective solution for safely treating Covid patients.In-vitro and in-silico studies conducted by ReaGene Innovations Private Limited, a startup company incubated at the ASPIRE-BioNEST, and INDRAS Private Limited, indicate that re-purposing the anti-diabetic drug Ertugliflozin, might provide a therapeutic solution to the Covid-19 infection.Ertugliflozin, an FDA approved drug for type-2 diabetes, works as SGLT-2 inhibitor by removing excessive glucose through urine.The research findings indicate that this re-purposed drug not only binds effectively to the receptor binding domain of the spike protein of Covid-19 and further blocks binding to human ACE2 but also displays significant anti-inflammatory and anti-thrombotic properties in a 3D human vascular lung model, both of which are fundamentals in Covid infection.This is the first result that offers a safe, ready-to-use, cost-effective solution to humans who contract Covid."It has immense potential to treat Covid infection, and our research proves its efficacy in the test-tube assays", said Dr Uday Saxena, CEO of ReaGene Innovations, co-founded with Dr Subramanyam Vangala, and Dr Sreedhara Voleti, MD of INDRAS.INDRAS focus is on consulting, contracting, and collaborative solutions to in-silico drug design, has prioritized about 8,000 FDA approved drugs to top-10 from their computational studies, which were further experimented by ReaGene Innovations for various in-vitro assays on cytokine storm, antithrombotic properties, and inflammatory marker reduction through various in vitro assays.The path to find such a re-purposed drug was critically planned and completed within a year of funding from the IT giant, Tech Mahindra.The outcomes of the results of this research were recently published in a journal (BioRxIV) and a joint patent has been filed both in India as well as internationally under Patent Cooperation Treaty.The results obtained are highly encouraging, and further in animal models towards preclinical, and clinical outcomes in humans are yet to be conducted for this drug to be officially nominated as a therapeutic agent for COVID-19, the company said in a statement.ASPIRE-BioNEST is a life sciences incubator jointly funded by BIRAC, a section-8 not-for-profit organization of Department of Biotechnology, Govt of India, and University of Hyderabad (UoH) for nurturing scaling technologies through entrepreneurship.--IANSpvn/skp/

Chennai, June 25 (IANS) The Defense Research and Development Organisation (DRDO) on Friday told the Madras High Court that 40 pharmaceutical companies have evinced interest in the anti-Covid-19 drug produced by it.The submission came in response to a Public Interest Litigation complaining that DRDO had given a license to Dr Reddy's Laboratories in Hyderabad for the production of the anti-Covid-19 drug.The premier scientific organisation said that it has invited Expressions of Interest (EoIs) to produce its D-glucose (2-DG) and adjunct drug to treat Covid patients, and already 40 pharmaceutical companies had expressed interest.Additional Solicitor General R. Sankaranarayanan told a bench of Justices N. Kirubakaran and T.V. Thamilselvi that the DRDO was interested in ensuring that the drug, developed by its Institute of Nuclear Medicine and Allied Sciences, could be manufactured in as many quantity as possible in the shortest possible time.The PIL filed by Chennai resident, D. Saravanan, complained that the DRDO had given license to Dr Reddy's Laboratories in Hyderabad alone for making the oral powder at a cost of Rs 990 for a sachet of 2.34 grams. The court also took note of a submission made by a litigant that the DRDO's medicine was very effective and a woman Covid-19 patient with comorbidities had a magical cure after being administered with 30 sachets of the drug at a cost of Rs 30,000. The litigant said that the doctor who had treated the patient had said that he can vouch for its efficiency.Sankaranarayanan told the court that that the DRDO had fixed June 17 as the last date for submission of EoIs and that 40 pharmaceutical companies had envisaged interest to manufacture the medicine with the DRDO knowhow. Washington, June 18 (IANS) The Biden administration has announced plans to invest more than $3 billion to accelerate the discovery, development and manufacturing of antiviral drugs to combat Covid-19.The Antiviral Program for Pandemics will respond to the urgent need for antivirals to treat Covid-19 by spurring the availability of medicines to prevent serious illness and save lives, the Department of Health and Human Services, said in a statement on Thursday.The programme aims to develop highly effective oral antiviral medicines that can be taken at home early in the course of infection, similar to antiviral treatment for influenza. This would prevent overwhelming surges in hospitalisations and save lives. This plan will support research to identify and accelerate availability of breakthrough treatment options to ensure the public has access to viable medicines.It also will build sustainable platforms for discovery and development of antivirals for other viruses with pandemic potential, helping better prepare the nation to face future viral threats."New antivirals that prevent serious Covid-19 illness and death, especially oral drugs that could be taken at home early in the course of disease, would be powerful tools for battling the pandemic and saving lives," said Dr Anthony Fauci, chief medical adviser to the President and Director of National Institute of Allergy and Infectious Diseases (NIAID), in the statement.As part of the plan, the National Institutes of Health (NIH) will evaluate, prioritise and advance antiviral candidates to Phase 2 clinical trials. The plan provides more than $300 million for research and lab support, nearly $1 billion for preclinical and clinical evaluation, and nearly $700 million for development and manufacturing through NIAID and the Biomedical Advanced Research and Development Authority (BARDA).Additionally, the plan allocates up to $1.2 billion to support the creation of collaborative drug discovery groups called Antiviral Drug Discovery (AViDD) Centers for Pathogens of Pandemic Concern that will harness the creativity of the biomedical research community and drive innovative antiviral drug discovery and development.These centres will create platforms that will initially target coronaviruses, and then could be expanded to other viruses with pandemic potential - helping to better prepare the nation for future viral threats.--IANSrvt/vd
